Viruses face selective pressures, but some inefficiencies can be masked. @jkpfeiff &co reveal an inefficiency in #norovirus exit from cells that can be overcome by a mutation in the virus NS3 protein, enhancing #CellDeath & progeny release @PLOSBiology plos.io/3ZQIDqi

Non-enveloped viruses are released from cells through several mechanisms, including lytic and non-lytic pathways. For murine norovirus, efficient egress relies on induction of cell death by the viral NS3 protein through interaction with mitochondrial lipids and subsequent cell lysis. Here, Rodriguez Irizarry et al. performed a forward genetic screen to identify murine norovirus mutants with enhanced replication speed. They show that a mutation in NS3 enhances viral egress through increased targeting of mitochondrial lipids and earlier induction of cell death. These results suggest that viral egress is an inefficient step of the replication cycle that can be overcome through an adaptive mutation, highlighting the importance of NS3-mediated viral egress.
