Metformin shown to prevent Long COVID across risk groups in multiple randomized trials

MINNEAPOLIS/ST. PAUL (1/29/2026) — Multiple randomized clinical trials and electronic health record studies now show that metformin, a widely used and well-established medication, significantly reduces the risk of developing Long COVID when taken during or shortly after acute infection with SARS-CoV-2. The findings were published today in Clinical Infectious DiseasesExternal link that opens in the same window.

In the paper, the University of Minnesota Medical School research team provides information about metformin and summarizes several studies in low, standard and high risk populations that show taking metformin during or soon after infection with SARS-CoV-2 prevents the development of Long COVID. Notably, the research summarizes the number of patients needed to receive treatment in each study to prevent one case of Long COVID. On average across the four studies, one case of Long COVID is prevented for every 50 cases of acute SARS-CoV-2 treated with 14 days of metformin.

Wastewater surveillance reveals true scale of COVID-19 spread as clinical testing declines

Osaka, Japan - As COVID-19 testing becomes less routine, official case numbers can make outbreaks look smaller than they really are. A research team led by Professor Michio Murakami has now shown that wastewater surveillance can uncover this “invisible” spread, providing a more objective picture of community infections and offering early warning signs for hospital-acquired cases.

Traditionally, public health authorities have relied on data from clinical tests, such as PCR, to monitor infectious disease outbreaks. However, this approach has limitations. When the number of tests performed decreases. For instance, after a change in public health policy or a drop in public concern, the official statistics may no longer reflect the true prevalence of the virus. New research shows that after the body’s defenses kill the virus behind COVID-19, leftover digested chunks of SARS-CoV-2 spike protein can target specific immune cells based on their shape. The revelations could explain why certain populations of cells that detect and fight infection are depleted in patients with severe COVID-19, and shed light on the omicron variant’s milder symptoms.

The study, published in the Proceedings of the National Academy of Sciences, may launch a line of inquiry that informs new strategies for quelling the most serious symptoms of COVID-19. Led by a UCLA team, the scientific collaboration comprises nearly three dozen engineers, microbiologists, immunologists, chemists, physicists, medical researchers and analytical experts.

On the basis of the weighted samples, we estimated 1,538,000 (1.4%) school-aged children had long COVID at some point." "69.7% were 12–17 years of age, 59.4% were female and 40.6% were male, and 35.2% were experiencing long COVID symptoms at time of survey." "Compared with children who never had long COVID, children who had long COVID had a higher prevalence of functional limitations across 5 of the 6 functional domains."

"The fact that average elevation in pTau-181 grew over the observational period may, if replicated longitudinally, suggest a temporal lag between COVID-19 onset and increases in circulating levels of tau phosphorylation."
